Resetting the Eye’s Cellular Clock: First Human Trial Results

The first safety test is done, and it is good.

Life Biosciences has released the first human Phase 1 clinical trial safety data for ER-100, an AAV2 gene therapy testing partial epigenetic reprogramming in patients with open-angle glaucoma. In this video, we analyze the 56-day safety profile, early visual field perimetry results, and how inducible OSK transcription factors aim to restore optic nerve function.

For nearly two decades, scientists have explored whether Yamanaka factor cellular reprogramming could safely operate in living humans. Now, Life Biosciences has reported the first-in-human Phase 1 interim safety readout for ER-100, an AAV2-delivered gene therapy targeting retinal ganglion cells in patients with primary open-angle glaucoma.

In this video, we review the 56-day safety data across the initial 3-patient cohort, which demonstrated zero serious adverse events and unanimous Data Safety Monitoring Board approval to advance to a 3-fold higher dose. We break down the early Humphrey visual field perimetry results showing a 4-decibel gain across 28% of test points in 2 patients, while contextualizing the clinical noise and learning effects common in glaucoma testing. Finally, we explore why the eye is the ideal testing ground for gene therapy, how ER-100 uses an oral doxycycline switch to drive OSK without the c-Myc oncogene, and what preclinical work from Dr. David Sinclair’s Harvard lab reveals about reversing vision loss.
