Emerging treatments for autoimmune disorders focus on \u201cadoptive cell therapies,\u201d or those using cells from a patient\u2019s own body to achieve immunosuppression. These therapeutic cells are recognized by the patient\u2019s body as \u201cself,\u201d therefore limiting side effects, and are specifically engineered to localize the intended therapeutic effect.<\/p>\n<p>In treating <a href=\"https:\/\/phys.org\/tags\/autoimmune+diseases\/\" rel=\"tag\" class=\"\">autoimmune diseases<\/a>, current adoptive cell therapies have largely centered around the regulatory T cell (T<sub>reg<\/sub>), which is defined by the expression of the Forkhead box protein 3, orFoxp3. Although T<sub>regs<\/sub> offer great potential, using them for therapeutic purposes remains a major challenge. In particular, current delivery methods result in inefficient engineering of T cells.<\/p>\n<p>T<sub>regs<\/sub> only compose approximately 5%\u201310% of circulating peripheral blood <a href=\"https:\/\/phys.org\/tags\/mononuclear+cells\/\" rel=\"tag\" class=\"\">mononuclear cells<\/a>. Furthermore, T<sub>regs<\/sub> lack more specific surface markers that differentiate them from other T cell populations. These hurdles make it difficult to harvest, purify and grow T<sub>regs<\/sub> to therapeutically relevant numbers. Although there are additional tissue-resident T<sub>regs<\/sub> in non-lymphoid organs such as in <a href=\"https:\/\/phys.org\/tags\/skeletal+muscle\/\" rel=\"tag\" class=\"\">skeletal muscle<\/a> and visceral adipose tissue, these T<sub>regs<\/sub> are severely inaccessible and low in number.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Autoimmune disorders are among the most prevalent chronic diseases across the globe.
