When it comes to delivering drugs to the body, a major challenge is ensuring that they remain in the area they’re treating and continuing to deliver their payload accurately. While major strides have been made in delivering drugs, monitoring them is a challenge that often requires invasive procedures like biopsies.

Researchers at NYU Tandon led by Jin Kim Montclare, Professor of Chemical and Biomolecular Engineering, have developed proteins that can assemble themselves into fibers to be used as therapeutic agents for the potential treatments of multiple diseases.

These biomaterials can encapsulate and deliver therapeutics for a host of diseases. But while Montclare’s lab has long worked on producing these materials, there was once a challenge that was hard to overcome—how to make sure that these proteins continued to deliver their therapeutics at the correct location in the body for the necessary amount of time.

A new study in Nature reports an AI-driven advance in biotechnology with implications for drug development, disease detection, and environmental monitoring. Scientists at the Institute for Protein Design at the University of Washington School of Medicine used software to create protein molecules that bind with exceptionally high affinity and specificity to a variety of challenging biomarkers, including human hormones.

Notably, the scientists achieved the highest interaction strength ever reported between a computer-generated biomolecule and its target.

Senior author David Baker, professor of biochemistry at UW Medicine and Howard Hughes Medical Institute investigator, emphasized the potential impact: “The ability to generate novel proteins with such high binding affinity and specificity opens up a world of possibilities, from new disease treatments to advanced diagnostics.”

The background: In a study published in 2020, researchers at Tufts University and the University of Vermont (UVM) harvested and incubated skin cells from frog embryos until they were tiny balls.

They then sculpted the spheres into specific shapes — dictated by an algorithm — and added layers of cardiac stem cells to them in precise locations.
